Description

UAS regulatory sequences drive expression of Mmus\Numb coding sequences where the entire C-terminus of Mmus\Numb (amino acids 396-593), which also contains the SH3 binding motifs is removed.

Expression of Mmus\NumbΔC.Scer\UAS in the SOP lineage, under the control of Scer\GAL4sca-537.4 or Scer\GAL4sca-109-68 causes balding of the notum due to transformation of IIA daughter into a second IIB and subsequent transformation of IIIb's sheath daughter into another neuron, resulting in mutant external sensory organs with as many as four neurons.

Co-expression of Mmus\NumbΔC.Scer\UAS with numbdsRNA.Scer\UAS.T:Hsap\MYC, both under the control of Scer\GAL4sca-537.4 rescues the numbdsRNA.Scer\UAS.T:Hsap\MYC neuronal-loss phenotype, with the abdominal segments, in particular a1, a3, a5, and a7, no longer exhibiting the neuronal loss found in numbdsRNA.Scer\UAS.T:Hsap\MYC mutants. This rescue of neuron and sheath cells is also observed in other regions of the PNS, including the lateral chordotonal organs.
