Comment:

Deletion resulting from the imprecise excision of P{lacW}LamP. The downstream deletion boundary within exon 3 of Lam was estimated from the map in Fig. 2.

Five day-old LamD395/LamK2 mutant flies exhibit severe midgut hyperplasia, equivalent to that seen in 50 day-old wild type flies.

The eclosion rate of LamD395 mutant flies is increased when the flies are raised in axenic conditions.

Suppressing secretion in fat body cells expressing Scer\GAL4Cg.PA results in a reduction in gut hyperplasia in LamD395/LamK2 mutant flies.

Most gonads of LamD395/Df(2L)cl-h1 male larvae are small, being filled with early germ cells and almost completely lacking differentiated spermatocytes and spermatids. The mutant early germ cells contain spectrosome-like structures and poorly branched fusome-like structures. Only 2.4% of the mutant His3-positive cysts contain 4 or 8 germ cells (in contrast to 52.2% of His3-positive cysts in wild type) and the cysts are distributed throughout the gonad. The nuclei of many cyst stem cells in the mutant gonad are positioned adjacent to hub cells (in wild type the nuclei are dispersed away from the hub), while the number of germline stem cells associated with the hub is decreased compared to wild type. Cyst cells are displaced away from germ cells in the mutant gonad, with many being mislocalised to the surface.

LamD395/Df(2L)cl-h1 trans-heterozygotes exhibit less and later lethality than LamD395 homozygotes, suggesting the presence of other genetic aberrations.

LamD395/Df(2L)cl-h1 trans-heterozygous larvae move significantly less than controls or LamD395/+ heterozygotes.

LamD395 homozygous and heterozygous mutants exhibit lower pupariation height, indicative of reduced larval locomotion and negative gravitaxis.

Expression of keyGD1249 under the control of Scer\GAL4Cg.PA suppresses the increased intestinal hyperplasia seen in LamD395/LamK2 mutant flies.

Co-expression of PGRP-LBScer\UAS.T:Avic\GFP-YFP and PGRP-SC2Scer\UAS.cCa under the control of Scer\GAL4Cg.PA suppresses the increased intestinal hyperplasia seen in LamD395/LamK2 mutant flies.

Expression of rlScer\UAS.T:Uuuu\nls6 under the control of Scer\GAL4tj-NP1624 rescues the defects in germline stem cell differentiation seen in LamD395/Df(2L)cl-h1 males. In 40.6% of testes spermatocytes are formed and the gonads do not accumulate many early germline cells. The mislocalisation of the cyst stem cell nuclei adjacent to the hub and the reduction in number of germline stem cells associated with the hub are also partially rescued.

LamK2/LamD395 is partially rescued by LamUAS.cMAa

Comments

Expression of LamScer\UAS.cMAa in the fat body under the control of Scer\GAL4Cg.PA significantly rescues the intestinal hyperplasia seen in LamD395/LamK2 mutant flies. The eclosion rate is increased from ~8% to ~40%.
