Description

UAS drives expression of the GH12907 EndoA cDNA (in which lysine at position 129 has been changed to arginine in order to comply with the EndoA 'consensus sequence' in FlyBase), containing T63S, A66S and A70Q amino acid changes.

Expression of endoASSQ.Scer\UAS using Scer\GAL4elav-C155 rescues endoAΔ4 larval lethality to adulthood, although the adult rescuants cannot fly and have reduced locomotor activity, and they have a median survival time of only 2.3 days. This genotype does rescue the severe tetanus-induced depression in excitatory junctional potentials (EJPs), the slow post-tetanic recovery of EJP amplitude, and the low frequency of spontaneous miniature EJPs seen in endoAΔ4 mutants.
