Flies show expressing tefudsRNA.Scer\UAS under the control of Scer\GAL4repo reduced climbing ability compared to wild type. In a countercurrent assay, 70% of the mutant flies are poor climbers, 16% moderate climbers and only 14% are good climbers (compared to wild-type values of 13% poor/moderate climbers and 87% good climbers). Flies which are poor climbers have significantly shorter longevity than good climbers, with the moderate climbers having intermediate longevity, suggesting that the difference in climbing ability between individual flies is due to differences in the severity of neurodegeneration.
Flies expressing tefudsRNA.Scer\UAS in glial cells under the control of Scer\GAL4repo exhibit climbing defects. They also exhibit significantly reduced longevity compared to controls, and unlike in wild type, holes are seen in the adult lamina and neuropil. Significantly more apoptotic (Caspase 3 positive) cells are seen than in wild type. At 7 days these are limited to neurons, but also include glial cells at 17 days. Mobility, longevity, brain morphology of apoptosis levels are not adversely affected when tefudsRNA.Scer\UAS is expressed in neurons under the control of Scer\GAL4elav.PLu, and flies have increased lifespan compared to controls. Lifespan and apoptosis levels are not affected when tefudsRNA.Scer\UAS is expressed using Scer\GAL4elav-C155, but a slight reduction in climbing ability is observed in young flies. No holes are seen in the brain, but several round structures with a darkly stained interior core are observed in the optic lobe.
Expression of tefudsRNA.Scer\UAS under the control of Scer\GAL4elav.PU results in neurodegeneration phenotypes. Swelling, fragmentation and loss of rhabdomeres in 3-day old adult eyes is observed.
Expression of tefudsRNA.Scer\UAS under the control of Scer\GAL4GMR.PU results in neurodegeneration phenotypes. Swelling, fragmentation and loss of rhabdomeres in 3-day old adult eyes is observed, and the extent of neurodegeneration becomes more severe with age. Eyes from these animals show a rough-eye phenotype.
Expression of tefudsRNA.Scer\UAS under the control of Scer\GAL4hs.PB during development (animals heat-shocked twice daily from egg-laying until day 3), or during adulthood (adults heat-shocked twice daily from day 3 to day 15), both result in degeneration of adult photoreceptor neurons.
Dividing photoreceptor neurons are observed posterior to the morphogenetic furrow in third instar larval eye discs expressing tefudsRNA.Scer\UAS under the control of Scer\GAL4GMR.PU or Scer\GAL4elav.PU.
Reduced cell death directly posterior to the morphogenetic furrow, and increased cell death more posterior to the morphogenetic furrow is observed in third instar larval eye discs expressing tefudsRNA.Scer\UAS under the control of Scer\GAL4GMR.PU. A small number of these dying cells are neurons.

stgEY12388/+ suppresses the rough-eye phenotype and increases the number of normal photoreceptors per ommatidium in flies expressing tefudsRNA.Scer\UAS under the control of Scer\GAL4GMR.PU. Significantly reduced severity of progressive neurodegeneration is also observed in these animals.
Rpd304556/+ suppresses the rough-eye phenotype and increases the number of normal photoreceptors per ommatidium in flies expressing tefudsRNA.Scer\UAS under the control of Scer\GAL4GMR.PU.

Expression of tefudsRNA.Scer\UAS under the control of Scer\GAL4elav.PU results in increased levels of photoreceptor cell division in third instar larval eye discs, which is suppressed by stgEY12388/+.
