Use of Scer\GAL80ChAT.3.3P restricts expression of a UAS target to the CLI1 and CLI2 neurons by inhibiting Scer\GAL4GMR47E12 expression in other interneurons. Use of Scer\GAL80tsh-GAL80 restricts expression of a UAS target to the CLI1 neurons by inhibiting Scer\GAL4GMR47E12 expression in other interneurons.
Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 660 nm (red) light) of a subset of Scer\GAL4GMR47E12 cells (expression restricted using Scer\GAL80ChAT.3.3P, or the combination of Scer\GAL80ChAT.3.3P and Scer\GAL80tsh-GAL80) halts forwards locomotion and significantly induces contraction of the abdomen, compared to controls. In the case of the subset of Scer\GAL4GMR47E12 cells activated when only Scer\GAL80ChAT.3.3P is also present, neuromere-specific activation results in segment-specific contraction of body wall muscles.
