Embryos expressing Hsap\BCL2Scer\UAS.cGa ubiquitously under the control of Scer\GAL4da.G32 fail to exhibit most of the cell death pattern normally seen during mid-embryonic development. Cell death appears particularly reduced in the lateral part of the cephalic region as well as in the ventral portion of the embryo during germ band retraction. The number of dying cells throughout the central nervous system is also reduced as the ventral nerve cord condenses.

Hsap\BCL2Scer\UAS.cGa heterozygotes are sub-viable, for all lines tested. Upon induction with Scer\GAL4da.G32, the percentage of lethality increases from 25% to 37%. Lethality occurs at earlier stages upon increasing the dose of Scer\GAL4da.G32 or Hsap\BCL2Scer\UAS.cGa, ranging from mostly larval death to embryonic death.

Ubiquitous expression of Hsap\BCL2Scer\UAS.cGa (under the control of Scer\GAL4da.G32 significantly reduced cell death induced by X-rays (4000 Rads) and accompanying levels of lethality.

Hsap\BCL2Scer\UAS.cGa expression in the developing eye disc (under the control of Scer\GAL4GMR.PF) leads to a rough eye phenotype in 45% of flies. This phenotype is enhanced and affects 100% of the flies expressing two doses of the Hsap\BCL2Scer\UAS.cGa transgene under the control of Scer\GAL4GMR.PF. These flies also exhibit supernumerary ommatidia and larger ommatidia, compared to wild-type. The retinal cell layer is disorganized compared to the regular pattern in wild-type. An increased spacing corresponding to additional cellular material between ommatidia is observed next to R1-R6 for each ommatidial unit. The position of this extra material correlates with the position where some of the pigment cells normally die by apoptosis in the pupal retina. A normal complement of photoreceptor cells is observed, indicating that Hsap\BCL2 alters the organisation of the eye by inhibiting apoptosis of pigment cells in the developing eye.

Expression of Hsap\BCL2Scer\UAS.cGa in the wing disc under the control of Scer\GAL4vg.PM has no detectable effect.

Expression of Hsap\BCL2Scer\UAS.cGa partially suppresses the rough eye phenotype seen in flies expressing Hsap\DEKScer\UAS.cLa under the control of Scer\GAL4GMR.PU.

Ubiquitous co-expression of Hsap\BCL2Scer\UAS.cGa and Mmus\BaxScer\UAS.cGb under the control of Scer\GAL4GMR.PF results in a pattern of cell death similar to flies expressing Hsap\BCL2Scer\UAS.cGa alone.

Expression of Hsap\BCL2Scer\UAS.cGa is able to rescue the lethality of Mmus\BaxScer\UAS.cGb-expressing flies at 21[o]C (both under the control of Scer\GAL4vg.PM).

Co-expression of Hsap\BCL2Scer\UAS.cGa in Mmus\BaxScer\UAS.cGb-expressing flies at 18[o]C induces a decrease in the ratio of strong wing phenotypes as opposed to intermediate and weak phenotypes and an increase of the ratio of weak wing phenotypes as opposed to intermediate and strong phenotypes. No significant effect was observed at 21 and 25[o]C. The structure of the wing is partially restored in these double mutants.
