Expressing aPKCHMS01320 under the control of Scer\GAL4wor.PA leads to defects in cell polarity in larval neuroblasts.

The expression of aPKCHMS01320 under the control of Scer\GAL4insc-Mz1407 leads to type II neuroblasts in the third instar larval brain abnormally exhibiting symmetric cell divisions, and presenting significant decreases in cell size and number per brain lobe, as compared to controls.

Embryos from mothers expressing aPKCHMS01320 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4otu.T:Hsim\VP16 or under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 display abnormal centrosome positioning in the embryonic epithelium.

Females heterozygous for aPKCHMS01320 and MTD-Gal4 (consists of Scer\GAL4otu.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4nos.UTR.T:Hsim\VP16) crossed to aPKCHMS01320 males results in: "few eggs,28% hatching"; maternal effect.

Testes of males expressing aPKCHMS01320 under the control of the triple "MTD" driver line (Scer\GAL4nos.PG, Scer\GAL4nos.UTR.T:Hsim\VP16 and Scer\GAL4otu.T:Hsim\VP16) sometimes contain misoriented spermatid cysts where the clustered spermatid nuclei are found apically (rather than basally).

The decreased number of type II neuroblasts in third instar larval brain lobes resulting from the expression of aPKCHMS01320 under the control of Scer\GAL4insc-Mz1407 is fully suppressed by the combined co-expression at low levels of Su(Tpl)Scer\UAS.SR.T:Hsap\MYC and lilliScer\UAS.SR.T:Zzzz\FLAG.

Co-expression of bazdsRNA.shRNA.Scer\UAS.P\T rescues the abnormal centrosome positioning of embryos from mothers expressing aPKCHMS01320 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4otu.T:Hsim\VP16.

par-1W3/+, or co-expression of Dhc64CGL00543, Dhc64CHMS01587, but not bazS980E.Scer\UAS.P\T.T:Avic\GFP-m6 or bazC.Scer\UAS.P\T.T:Avic\GFP-EGFP, rescues the abnormal centrosome positioning of embryos from mothers expressing aPKCHMS01320 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16.
