The co-expression of Su(Tpl)Scer\UAS.SR.T:Hsap\MYC and lilliScer\UAS.SR.T:Zzzz\FLAG in third instar larval brains leads to a significant increase in the number of type II neuroblasts as compared to controls under the following conditions: constitutive or clonal co-expression under the control of Scer\GAL4pnt-14-94; constitutive co-expression at high levels, but not at low levels, under the control of Scer\GAL4insc-Mz1407. The neuroblasts derived from Scer\GAL4pnt-14-94-driven co-expression display normal cortical polarity but are highly proliferative, giving rise to an enlargement of the brain, and their transplantation into the abdomen of wild-type adults leads to highly proliferative overgrowths that metastasize to distant tissues, as compared to controls.

The increased number of type II neuroblasts resulting from Scer\GAL4pnt-14-94-driven co-expression of Su(Tpl)Scer\UAS.SR.T:Hsap\MYC and lilliScer\UAS.SR.T:Zzzz\FLAG is enhanced by one copy of E(spl)mγ-HLHT:Avic\GFP and partially suppressed by one copy of Df(3R)grob32.2; the increased number of type II neuroblast resulting from Scer\GAL4insc-Mz1407-driven co-expression is enhanced by the additional co-expression of Su(H)Scer\UAS.T:SV5\V5, is fully suppressed by the additional co-expression of Cdk9D199N.Scer\UAS.SR and is even converted into a near absence of type II neuroblasts as compared to controls by the additional co-expression of either NGD144 (together with Dicer-2 transgene, for efficient RNAi) or DlD.Scer\UAS.

The co-expression of Su(Tpl)Scer\UAS.SR.T:Hsap\MYC and lilliScer\UAS.SR.T:Zzzz\FLAG under the control of Scer\GAL4ase.neuro does not lead to significant changes in the number of type I neuroblasts in the third instar larval brain, as compared to controls.

The combined co-expression at low levels of Su(Tpl)Scer\UAS.SR.T:Hsap\MYC and lilliScer\UAS.SR.T:Zzzz\FLAG fully suppresses the decreased number of type II neuroblasts in third instar larval brain lobes resulting from the expression of aPKCHMS01320 under the control of Scer\GAL4insc-Mz1407.
