UASt regulatory sequences drive expression of a mutated form of pbl-RA cDNA (amino acids 386-853) that lacks the N-terminal regulatory domain, with a single amino replacement (V531D) in the DH domain (this mutation has been shown to compromise catalytic activity). The coding sequence is tagged at the C-terminal end with Tag:HA.
Scer\GAL4e22c-mediated expression of pblΔN-term-V531D.Scer\UAS.T:Ivir\HA1 results in 0.9% embryonic lethality with 0% of embryos showing some loss of naked cuticle on the ventral side.
Expression of pblΔN-term-V531D.Scer\UAS.T:Ivir\HA1 in the mesoderm under the control of the Scer\GAL4twi.2PE and Scer\GAL4Mef2.PR drivers does not produce any mesodermal invagination or migration defects.
