lexAop regulatory sequences drive expression of the spGFP11 split fluorescent protein fragment fused to a membrane targeting sequence (Tag:M(hCD4)). This construct is designed for use in a GRASP (GFP Reconstitution Across Synaptic Partners) assay, in which complementary split fluorescent protein fragments expressed on the extracellular membrane of different neurons can reconstitute a functional fluorescent protein at points of contact between the neurons.
Avic\GFPspGFP11.Ecol\lexAop.T:Hsap\CD4 forms one half of the split-GFP "GFP reconstitution across synaptic partners" (GRASP) system (see PMID:18255029). In this system two split-GFP halves are expressed on the membrane of distinct neuronal populations. Avic\GFP is reconstituted trans-synaptically and fluorescence is seen when the two neuron populations connect.
