Death of Caf1-180270 hemizygous males occur progressively over the three larval instars, but predominantly during the second instar. Generally, Caf1-180270 mutant larvae are developmentally arrested one or two days prior to their death. Rarely, a few escapers can develop into third instar larvae where they persist up to the 18th day of life. Such arrested larvae appear normal but remain smaller than the wild-type and display a smaller fat body.

Females carrying Caf1-180270 mutant germline clones are sterile. The ovaries with Caf1-180270 mutant germline clones fail to develop beyond stage 10 of oogenesis.

Flies with Caf1-180270 mutant clones induced in their eyes display tissue specific abnormalities. While some flies lack eyes completely, others exhibit extremely small eyes with disorganized ommatidia.

Heterozygous Caf1-180270 suppresses the eye colour and bristle position effect variegation (PEV) phenotypes of In(2)Heidi-P{Winkelried}D and T(2;3)SbV, respectively.

Heterozygous Caf1-180270 mutants are more sensitive to gamma-irradiation that is known to lead to double-strand DNA breaks. The efficiency of homologous recombination gap repair is also reduced in Caf1-180270 heterozygotes, although not completely abolished.

Comments

Ubiquitous expression of Caf1-180Scer\UAS.cSa under the control of Scer\GAL4ptc-559.1 completely rescues the mutant phenotypes associated with Caf1-180270.

Caf1-180Scer\FRT rescues the lethality and sterility of Caf1-180270. The rescued animals do not show any visible defects.
