UASt regulatory sequences drive expression of a 500bp inverted repeat.
lethal, with Scer\GAL4c179
viable, with Scer\GAL4G7
Expression of iclnNIG.4924R under the control of any of the following: Scer\GAL4Mef2.PR, Scer\GAL4RapGAP1-OK6 (both with Dcr-2Scer\UAS.cUa), Scer\GAL4c179, Scer\GAL4how-24B, Scer\GAL4G7 or Scer\GAL4da.PU (all without Dcr-2Scer\UAS.cUa) reduces the adult viability to a varying degree (up to a point of complete lethality in some cases) at at least one of these two temperatures: 29[o]C and 25[o]C (with more severe effects being observed at 29[o]C). Expression under Scer\GAL4Mef2.PR, Scer\GAL4RapGAP1-OK6 (both without Dcr-2Scer\UAS.cUa) or Scer\GAL4elav.PU (with or without Dcr-2Scer\UAS.cUa) does not significantly decrease adult viability at either temperature.
Expression of iclnNIG.4924R (together with Dcr-2Scer\UAS.cUa to enhance RNAi efficiency) under the control of Scer\GAL4Mef2.PR results in severe flight defects with most flies being completely flightless, the adult flies also display significant drop in locomotor activity both during day and at night compared to controls.
iclnNIG.4924R is rescued by iclnUAS.cBa/Scer\GAL4Mef2.PR
iclnNIG.4924R is partially rescued by iclnUAS.cBa/Scer\GAL4da.PU
The lethality of flies expressing iclnNIG.4924R under the control of Scer\GAL4da.PU and the flightless phenotype observed in flies expressing iclnScer\UAS.cBa under the Scer\GAL4Mef2.PR driver is partially rescued and fully rescued, respectively, by co-expression of iclnScer\UAS.cBa.
