Larval wing disc clones expressing eddsRNA.Scer\UAS.intra sort out from the surrounding wild type cells, exhibiting both rounded and smooth contours rather than the jagged border seen in controls. The wild type cells surrounding the clone assemble a ring of actomyosin cable at the interface and the clone cells themselves develop an apical constriction that is inversely proportional to the size of the clone.

edΔintra.Scer\UAS.T:Zzzz\FLAG twinspot clones generated in the dorsal compartment of larval wing discs expressing eddsRNA.Scer\UAS.intra under the control of Scer\GAL4ap-md544 are not surrounded by a prominent actomyosin cable. The clone cells do sort out to some degree from the ed deficient neighbouring cells, but as in wild type the clonal border is jagged. However, the clone cells do show strong apical constriction.

Expression of fredScer\UAS.T:Zzzz\FLAG rescues the cell sorting phenotype seen in wing disc clones expressing eddsRNA.Scer\UAS.intra. The surrounding wild type cells no longer assemble an actomyosin cable and the boundary between the mutant clone and neighbouring wild type cells remains smooth.

Expression of ed::NrgScer\UAS.T:Zzzz\FLAG is unable to suppress the cell sorting phenotypes seen in larval wing disc clones expressing eddsRNA.Scer\UAS.intra. An actomyosin cable still forms around the clone and the boundary between the mutant clone and neighbouring wild type cells remains smooth.

Comments

Expression of edΔintra.Scer\UAS.T:Zzzz\FLAG rescues the cell sorting phenotypes seen in larval wing disc clones expressing eddsRNA.Scer\UAS.intra. The surrounding wild type cells no longer assemble an actomyosin cable and the boundary between the clone and the surrounding wild type cells is no longer smooth edged, instead appearing jagged as in wild type.
