Larvae expressing TrhdsRNA.Sym.Scer\UAS under the control of Scer\GAL4elav-C155 exhibit a reduced feeding rate compared to controls. There is no change in pupal size or time to pupariation, but there is a correlation between the reduced feeding rate and increased pupal survival.

Animals with reduced Trh through expression of TrhdsRNA.Sym.Scer\UAS under the control of Scer\GAL4elav.Switch.PO (included by 6mg/ml RU486) exhibit a feeding rate indistinguishable from controls, when fed with 5-HTP. Changes in 5-HT levels either by feeding 5-HTP or by reducing Trh levels has no effect on locomotion.

Decreased levels of Trh through expression of TrhdsRNA.Sym.Scer\UAS under the control of Scer\GAL4elav-C155 directly correlate with reduced feeding relative to controls. General motor activity is unaffected. Feeding 5-HTP to these flies results in a highly increased appetite, while locomotor control is unaffected.

There is a correlative increase in neuronal arborization with decreasing levels of neuronal 5-HT in animals expressing TrhdsRNA.Sym.Scer\UAS under the control of Scer\GAL4elav-C155. In general, the number of total varicosities increase with TrhdsRNA.Sym.Scer\UAS expression.

Analysis of the architecture of serotonergic fibers projecting to the proventriculus in Scer\GAL4elav.Switch.PO/TrhdsRNA.Sym.Scer\UAS animals, inducing neuronal expression of the transgene during the second instar stage, reveals no differences in arborization of fibers, varicosity number, number of large varicosities, or in varicosity area.

Co-expression of pledsRNA.Sym.Scer\UAS and TrhdsRNA.Sym.Scer\UAS under the control of Scer\GAL4elav-C155, does not affect larval feeding, unlike expression of either single transgene. When the same combination is placed under the control of either Scer\GAL4ple.PF or Scer\GAL4l.Trh.PK, feeding is reduced, consistent with knockdown of only DA or 5-HT synthesis, but not both. Gut fiber architecture is rescued to control levels by the presence of both pledsRNA.Sym.Scer\UAS and TrhdsRNA.Sym.Scer\UAS.
