Detailed Description

Adult flies expressing mir-9aScer\UAS.T:Ppyr\LUC under the control of Scer\GAL4Hk-UH3 show wing posture defects and are completely flightless. Their dorsal longitudinal muscles are severely disrupted with broken muscle fibers that appear hypercontracted and whole fascicles missing, the sarcomere is disorganized and no clear Z-discs are formed.

Expression of mir-9aScer\UAS.T:Ppyr\LUC under the control of Scer\GAL4sd-SG29.1 results in curved, spoon-shaped wings which are slightly folded. The distribution of hairs on the wing margin is not homogeneous.

The severe dorsal longitudinal muscle (DLM) structural defects observed in adult flies expressing mir-9aScer\UAS.T:Ppyr\LUC under the control of Scer\GAL4Hk-UH3 are suppressed by combination with Mhc2B. Similarly, the disrupted DLM morphology, along with the sarcomere disorganization and to a large extent also the flightlessness of mir-9aScer\UAS.T:Ppyr\LUC-expressing flies is rescued by co-expression of either upΔmir-9a.Scer\UAS.10a or upΔmir-9a.Scer\UAS.10b.

Comments

The inability to fly along with severe structural defects in their indirect flight muscles (broken muscle fibers, disorganized sarcomere) characteristic for flies expressing mir-9aScer\UAS.T:Ppyr\LUC under the control of Scer\GAL4Hk-UH3 is rescued by co-expression of mir-9asponge.Scer\UAS.T:Disc\RFP-mCherry.
