UAS regulatory sequences drive expression of wild-type DCTN1-p150 which is tagged with a Tag:HA epitope.
Adults expressing DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ple.PF exhibit a significant decrease in lifespan and a significant, progressive decrease in climbing performance (detected at day 45, but not day 5, post eclosion)but 45 days old adults do not display a significant decrease in spontaneous locomotor activity or significant changes in the number of PPM1/2, PPM3 or PPL1 neurons, as compared to controls.
The third instar larval neuromuscular junctions of individuals expressing DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Toll-6-D42 display a significant increase in the density of dense core vesicles in distal, but not proximal, terminal boutons, without significantly affecting their size, as compared to controls; the respective axonal dense core vesicles exhibit a significant increase in size, as compared to controls.
Neurotransmission across the third instar larval neuromuscular junction of individuals expressing DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4RapGAP1-OK6 presents a significant increase in the frequency of miniature excitatory junction potential, but does not display significant defects in miniature excitatory junction potential amplitude, paired-pulse ratio, excitatory junction potential amplitude or quantal content, as compared to controls.

The decreases in adult climbing performance and in lifespan exhibited upon the expression of DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ple.PF or exhibited by TBPHΔ23 heterozygotes are suppressed in individuals that simultaneously express DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ple.PF and are heterozygous for TBPHΔ23. The expression of DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ple.PF also partially suppresses the decreased number of PPL1 neurons in the adult brain, but not the spontaneous locomotion defects, presented by 45 days old TBPHΔ23 heterozygotes.
The increased size of axonal dense core vesicles and the increased density of dense core vesicles in distal terminal boutons at the third instar larval neuromuscular junction of individuals expressing DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Toll-6-D42 are suppressed and enhanced, respectively, by TBPHΔ23 heterozygosity.
The expression of DCTN1-p150Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Toll-6-D42 enhances and suppresses the increased density of dense core vesicles at the distal and proximal terminal boutons, respectively, of TBPHΔ23 heterozygous third instar larval neuromuscular junctions.
