Description

UASp regulatory sequences drive expression of the FH1 and FH2 domains of dia. The protein is tagged at the N-terminal end with EGFP (the tag is joined to the dia coding region via a HRYTSLYKKAGSAAAPFT linker).

Expression of diaFH1FH2.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4en-e16E results in the dynamic lamellipodia and filopodia of the leading edge cells being replaced by lamellipodia with ruffled edges and by fan-shaped lamellipodia with radial actin arrays not seen in wild-type lamellipodia. These lamellipodia are larger than in wild type. Filopodial number, length and lifetime are all significantly decreased compared to wild type. Many of the remaining "filopodia" arise from fan-shaped lamellipodia and do not grow substantially after their appearance, in contrast to wild-type filopodia.
