UAS regulatory sequences drive expression of a short inverted repeat.
Second instar larvae expressing crbHMS01842 under the control of either Scer\GAL4da.G32 or Scer\GAL4btl.PS show twisted tracheal tubes and lack of gas filling; Scer\GAL4da.G32-driven expression leads to reduced apical surfaces of tracheal tube cells, despite of no obvious defects in apico-basal polarity. Scer\GAL4btl.PS-driven expression leads to significantly smaller Vps35-positive vesicles in larval tracheal cells.
Embryos derived from females expressing crbHMS01842 in the germline under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 show severe chromosome defects like chromosome bridges and bent chromosomes. They also show defects in spindle microtubules such as monopolar spindles and fusion with one or more neighbouring spindles. Anaphase chromosomes show high levels of PH3 staining in all chromosomal regions compared with wild type.
