apn1 mutant embryos show an apparently normal trachea during embryonic stages, even when derived from mutant germline clones. apn1 second instar larvae show unusually twisted and uninflated tracheal tubes, mostly manifested in the posterior tracheal metameres 9 and 10; this includes multicellular tubes, autocellular and seamless tubes; electron micrographs reveal a disorganized lumen with "tongues" of cellular protrusions into the lumen, although the epicuticle, procuticle and the thickenings formed by the aECM appear similar to controls; there are no obvious defects in secretion, endocytosis or apico-basal polarity. The dorsal trunk length is significantly reduced, as revealed by measurements of the posterior metamere length, but no significant difference is observed in cell number within different metameres, as compared to controls; the long apical surface length and the apical cell surface are reduced, although the short apical length is not affected, as compared to controls; mutant larval tracheal cells exhibit significantly larger Vps35-positive vesicles, despite of unaffected Arl8-positive lysosomal vesicles and Golgin245-positive trans-Golgi vesicles, as compared to controls.
