Detailed Description

Arfip12/Arfip71 third instar larvae have significantly fewer boutons at the neuromuscular junction (muscle 4) compared to Arfip12/+ heterozygotes. Arfip12/Arfip71 larval NMJs are similar to wild type in terms ofRMP, mEJP, EJP and quantal content.

Newly eclosed homozygous escapers have no obvious neurological phenotypes and are able to fly and climb walls.

The average number of boutons at the neuromuscular junction is significantly reduced compared to controls in Arfip71/Df(3R)ED5518 and Arfip71/Arfip12 third instar larvae. The average number of boutons at the neuromuscular junction is also reduced in Arfip71/Arfip12 second instar larvae.

Axonal transport is normal in Arfip71/Arfip12 third instar larvae.

The Golgi appears smaller and more fragmented than normal in the ventral nerve cord of Arfip71/Arfip12 third instar larvae.

Arfip12/Arfip71 flies are sensitive to ethanol-induced sedation compared to wild-type controls.

garzΔ137/+;Arfip71/+ or garzΔ137/+;Arfip12/+ third instar larvae have significantly fewer boutons at the neuromuscular junction compared to Arfip71/+ heterozygotes. garzΔ137/+ does not enhance the reduction in bouton number seen in Arfip71/Arfip12 larvae.

Sec5E10/+;Arfip71/+ or Sec151/+;Arfip71/+ third instar larvae do not show a reduction in bouton number at the neuromuscular junction, compared to Sec5E10/+ or Sec151/+ heterozygotes.

Comments

Expression of ArfipScer\UAS.WT driven by Scer\GAL4RapGAP1-OK6 (motor-neuron specific) rescues the reduced bouton number seen at the neuromuscular junction of Arfip12/Arfip71 third instar larvae. Expression of ArfipGBD.Scer\UAS.T:Ivir\HA1 by either Scer\GAL4elav-C155 (pan-neuronal) or Scer\GAL4RapGAP1-OK6 does not rescue the reduced bouton number seen at the neuromuscular junction of Arfip12/Arfip71 third instar larvae.

Expression of ArfipScer\UAS.WT under the control of either Scer\GAL4elav-C155 or Scer\GAL4Rapgap1-OK6 rescues the reduction in bouton number at the neuromuscular junction seen in Arfip71/Arfip12 third instar larvae.

Expression of ArfipBAR.Scer\UAS.T:Ivir\HA1 under the control of either Scer\GAL4elav-C155 or Scer\GAL4Rapgap1-OK6 fails to rescue the reduction in bouton number at the neuromuscular junction seen in Arfip71/Arfip12 third instar larvae.

Expression of ArfipScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PU at 29[o]C rescues the sensitivity of Arfip12/Arfip71 flies to ethanol-induced sedation.
