UAS regulatory sequences drive expression of an inverse repeat targeting a region common to four out of five predicted isoforms of pyd. The 519bp fragment begins 2662bp after the start codon of pyd-RB.

Adult flies expressing pyd3.dsRNA.Scer\UAS under the control of Scer\GAL4GMR.PF exhibit ectopic thoracic bristles and a mildly rough eye. Inter-ommatidial precursor cell (IPC) patterning errors are observed at 41-42 hours after pupal formation (APF). A mild increase is seen in the total number of IPCs surrounding each ommatidium, especially around the bristles. The IPCs are poorly placed, resulting in breakdown of the hexagonal pattern to pentagonal or squared ommatidial arrays and the integrity of the 1[o]/1[o] interface is occasionally disrupted by an IPC that makes direct contact with the cone cells (referred to as cone-contact cells). Three cells are observed to share the 3[o] cell niche equally, where normally a single cell is found in this position. Similar defects are observed when pyd3.dsRNA.Scer\UAS is expressed predominantly in IPCs under the control of Scer\GAL454. Defects in IPC patterning are also seen when pyd3.dsRNA.Scer\UAS is expressed under the control of Scer\GAL4GMR.PF at 20h APF. As in wild type, local movements are seen but these are undirected and unproductive and the cells often fail to undergo intercalation to create single cell rows.

The number of cone cells in the ommatidia of flies expressing pyd3.dsRNA.Scer\UAS under the control of Scer\GAL4GMR.PF is occasionally altered, and incorrect contacts are often seen within a cone cell quartet. 8.1% of mutant flies fail to undergo the change from having anterior/posterior (A/P) apical contacts to the equatorial/polar (E/P) configuration that occurs in wild type ommatidia between 22 and 28h APF, with a subset of cone cells maintaining the immature A/P arrangement through to 42h APF.

Expression of pyd3.dsRNA.Scer\UAS under the control of Scer\GAL4sd-SG29.1 has no visible effect on the wing veins.
