Description

Endogenous Dscam1 exons 7-11, which include the 33 exon 9 alternatives, are replaced with a cDNA encoding exons 7-11 which contains a single variant (variant 9) of exon 9, thus reducing the number of potential Dscam1 ectodomain isoforms from 19,008 to 576. (x.y.z in the allele symbol refers to combinations of exon 4, 6 and 9 respectively.). A single FRT site is present in the intron between exon 16 and exon 17.

Mutant (homozygous or heterozygous over a null allele) self dendrites of class I and class III dendritic arborization neurons, which normally overlap, show significant avoidance.

Mutants only form a single mushroom body lobe.

Dscam576-isoform.x.y.8/Dscam576-isoform.x.y.9 animals show abnormal mushroom body morphology: one lobe is absent or is thinner is the majority of these mutants.

Dscam576-isoform.x.y.9/Dscam576-isoform.x.y.25 animals show abnormal mushroom body morphology: one lobe is absent or is thinner is the majority of these mutants.

Mutants have marked defects in the axonal branching patterns of posterior scutellar neurons.
