Description

The mir-989 hairpin has been deleted. A w+mW.hs marker which is flanked firstly by a pair of loxP sites and secondly (flanking the loxP sites) by a pair of inverted attP sites is present in place of the deleted sequence.

Detailed Description

Embryos derived from mir-989KO/Df(2R)Exel7130 embryos show a significant decrease in the number of primordial germ cells compared to embryos from control females.

Border cell migration is often delayed in the ovaries of homozygous, mir-989KO/Df(2R)50C-38 and mir-989KO/Df(2R)Exel7130 females.

Wild-type border cells migrate normally when germline cells are homozygous for mir-989KO in mosaic egg chambers. In contrast, migration is delayed when somatic cells including the border cell cluster are homozygous but the germline is wild type. Mixed border cell clusters containing both wild-type and homozygous cells show a delay in migration which is similar in magnitude to the delay seen when the border cell cluster consists entirely of homozygous cells.

Comments

Expression of mir-989Scer\UAS.cKa under the control of Scer\GAL4slbo.2.6 fails to rescue the delayed border cell migration seen in mir-989KO females.

Clonal expression of mir-989Scer\UAS.cKa under the control of Scer\GAL4Act.PU in the border cell cluster rescues the delayed border cell migration seen in mir-989KO females.
