The majority of animals expressing Mi-2Δ932-1158.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4da.G32 complete larval development at 25[o]C, but only 5% survive to adulthood. At 29[o]C no animals survive beyond the first or second larval instar.
The lethality caused by expression of Mi-2Δ932-1158.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4da.G32 is moderately enhanced by Mi-2f08103/+ and is strongly enhanced by Mi-24/+.
Polytene chromosomes in larvae expressing Mi-2Δ932-1158.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4da.G32 and shifted from 25 to 29[o]C at the end of the first larval instar are thinner than normal and have a disrupted banding pattern (both in fixed squash preparations and in live analysis).
Animals carrying Mi-2Δ932-1158.Scer\UAS.T:Zzzz\FLAG, Scer\GAL4da.G32 and Scer\GAL80ts.αTub84B and shifted from 18 to 29[o]C during the third larval instar (which inactivates the temperature sensitive Scer\GAL80ts.αTub84B allele, allowing expression of Mi-2Δ932-1158.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4da.G32) show a significant decrease in polytene chromosome volume and a modest increase in polytene chromosome compaction.
