UAS regulates expression of Syt1 containing D282N and D284N mutations in the C2A domain and D416N and D418N mutations in the C2B domain, thus preventing Ca[2+] binding both domains.

Scer\GAL4elav-C155-mediated expression of Syt1C2A*-C2B*.Scer\UAS in a Syt1N13/Syt1AD4 background results in spontaneous neurotransmitter release frequency (measured as mEJPs) at third instar NMJs far beyond the levels observed in controls.

Scer\GAL4elav-C155-mediated expression of Syt1C2A*-C2B*.Scer\UAS results in a dramatic increase in nerve-evoked excitatory junction potential (eEJP) amplitude without affecting asynchronous release or spontaneous fusion (as measured by mEJPs) at third instar NMJs at low (0.2mM) external Ca[2+], compared to controls.

There are no discernible synaptic abnormalities associated with over-expression of Syt1C2A*-C2B*.Scer\UAS, including no significant increase in the number of synaptic vesicles near active zones, compared to over-expression of Syt1C2A-C2B.Scer\UAS. There is also no significant change in the number of release sites between these two genotypes.

Scer\GAL4elav-C155-mediated expression of Syt1C2A*-C2B*.Scer\UAS increases vesicle release probability and lowers the Ca[2+] concentration required to trigger fusion, compared to expression of Syt1C2A-C2B.Scer\UAS.

Scer\GAL4elav-C155-mediated expression of Syt1C2A*-C2B*.Scer\UAS partially rescues the reduction in docked synaptic vesicles and vesicle density at type 1b boutons in Syt1N13/Syt1AD4 third instar larvae.
