A mutated copy of the Tpi locus (containing the amino acid replacement M80T) has been inserted into the attP site present in TpiattP. A single attR site remains 5' and a single loxP site remains 3' to the reconstituted Tpi locus.

TpiM80T/TpiM80T, TpiM80T/TpiR187A, TpiM80T/TpiR187L, TpiM80T/TpiR187S and TpiM80T/Tpisgk-js10 individuals are bang sensitive, whereas TpiM80T/TpiR187K individuals are not.

TpiM80T homozygotes are partially lethal; TpiM80T homozygous and TpiM80T/Tpisgk-js10 transheterozygous adults exhibit a significant decrease in lifespan and an increase in heat sensitivity, shown by the significant decrease in the time until paralysis after a temperature shift to 38[o]C, as compared to controls; TpiM80T/Tpisgk-js10 transheterozygous adults, but not TpiM80T homozygous adults, exhibit an increase in bang sensitivity, shown by a significant increase in recovery time from mechanical stress, as compared to controls.
