Insertion of a Mi{Hto-WP} element into intron 2 of the longest iab-8 isoforms ( 3R:16 ,898,163 (-) -Release 6). The hostile takeover (hto) exon in the Mi{Hto-WP} element splices to the downstream iab-8 exon 3 (bases 1252-1484 of the iab-8-RC transcript - RefSeq NR_048413), before splicing into abd-A. This would ordinarily result in a fusion protein being expressed under the control of Scer\UAS regulatory sequences in the Mi{Hto-WP} element, but in this case the fusion has no extended reading frame.
leg | ectopic, with Scer\GAL4ey.PU
wing | ectopic, with Scer\GAL4Bx-MS1096
Expression of one copy of iab-8LNP under the control of Scer\GAL4GMR.PU produces flies that have a mild rough eye phenotype that is mottled with some fused ommatidia near the center.
Flies expressing iab-8LNP under the control of Scer\GAL4ey.PU die as pupae or pharate adults with head capsule and antennae shifted toward a limb fate.
Flies expressing iab-8LNP under the control of Scer\GAL4Bx-MS1096 have halteres that are shifted towards a wing fate.
When iab-8LNP is expressed around the time of pupation (by heat induction of Scer\GAL4hs.PB-Ubi), ectopic pigmentation appears on the abdomen, indicating a conversion of anterior abdominal segments toward a posterior identity.
