UAS regulatory sequences drive expression of a full-length bs open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.
The expression of bsKK108659 under the control of Scer\GAL41151 affects the indirect flight muscles myofibrils (although correctly specified, the muscles show a low polymerized actin content), but not jump muscles, as compared to controls. Although indirect flight muscle myofibril diameter is comparable to controls during mid-pupal stage, it is severely decreased in adults, suggesting myofibrils fail to grow from pupae to adults; however, sarcomere length is normal in both pupae and adults; the myofibril growth defects is rescued by the co-expression of bsUAS.ORF.GW.Tag:HA, with the myofibrils becoming even greater in diameter than those of controls.

The co-expression of bsUAS.ORF.GW.Tag:HA significantly improves the indirect flight muscles defects (actin content and myofibril diameter), but not the flight defects, induced by the expression of bsKK108659 under the control of Scer\GAL41151; and the rescued myofibrils even become greater in diameter than those of controls.
