UAS regulatory sequences drive expression of a full-length wek open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.
Expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4elav.PU results in increased number of apoptotic (Dcp-1-positive) cells in pupal ventral nerve cord (VNC) and decrease in the number of eve-positive neurons both in embryos and in pupal VNC, as compared to controls. Scer\GAL4Myd88-NP6394-driven expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 also leads to a moderate loss of eve+ neurons and cells within the driver's expression domain (assayed using a nuclear histone-reporter) in the pupal abdominal VNC.

The decrease in the number of cells within the Scer\GAL4Myd88-NP6394 driver's expression domain in the pupal ventral nerve cord (assayed using a nuclear histone-reporter) observed upon expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 is exacerbated further by co-expression of Myd88VDRC.cUa.
The reduced level of cell death (decreased number of Dcp-1-positive cells) observed in the ventral nerve cord (VNC) of wekEX14/Df(2L)BSC690 and the increase in cell death induced by Scer\GAL4elav.PU-driven expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 can both be mitigated by combining the two manipulations together: The number of apoptotic cells in the VNC of wekEX14/Df(2L)BSC690 mutants expressing wekScer\UAS.ORF.GW.T:Ivir\HA1 is intermediate to each of the two perturbations and higher than in controls.
