Description

UAS regulatory sequences drive expression of a full-length wek open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.

Expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4elav.PU results in increased number of apoptotic (Dcp-1-positive) cells in pupal ventral nerve cord (VNC) and decrease in the number of eve-positive neurons both in embryos and in pupal VNC, as compared to controls. Scer\GAL4Myd88-NP6394-driven expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 also leads to a moderate loss of eve+ neurons and cells within the driver's expression domain (assayed using a nuclear histone-reporter) in the pupal abdominal VNC.

Expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4Bx-MS1096 results in male flies with severe wing size and patterning defects.

Expression of wekScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4Bx-MS1096 results in female flies with weak wing size and patterning defects.
