The FRT cassette present in the wgKO.FRT.NRT-wg.FRT.wg progenitor allele has been excised, removing 'Nrt-wg' and allowing the wild-type wg sequence to be expressed under the control of endogenous wg regulatory sequences. The loxP cassette (containing a Disc\RFPmCherry.3xP3 marker) present in the progenitor allele is still present downstream of the wg coding region.
"wg{KO; NRT-Wg[Body]; WT[Discs]}" larvae expressing wild-type wg in imaginal discs and 'NRT-wg' (a membrane-tethered form of wg) in other tissues have been generated by expressing FLPase in the imaginal discs of homozygous wgKO;FRT.NRT-wg.FRT.wg larvae (converting wgKO;FRT.NRT-wg.FRT.wg to wgKO;-FRT.wg in the discs). These animals largely fail to eclose when co-cultured with control siblings, but eclose into normal-looking adults when grown in separate vials.
