FlyBase curator comment: FBrf0246471 sequenced the ykt6[A] and ykt6[C] mutant allele stocks and reported that ykt6[A] carries the Q62R mutation (not M1I) and ykt6[C] carries the M1I mutation (not Q62R).

FBrf0246471 sequenced the ykt6[A] and ykt6[C] mutant allele stocks and reported that ykt6[A] carries the Q62R mutation (not M1I) and ykt6[C] carries the M1I mutation (not Q62R). Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change.
Eggs derived from Ykt6C germline clones show obvious structural defects, including and small round eggs, occasionally with nurse cells remnants still attached, absent, fused, shortened and broad dorsal appendages, Most severely affected egg chambers arrest early in oogenesis as a syncytium of nurse cell nuclei sharing a common cytoplasm. Intermediate stages of oogenesis exhibit a plethora of defects: multinucleate nurse cells show loss of plasma membrane, aggregates of actin and ring canals, with the ring canals exhibiting excessive associated F-actin, plasma membrane fragments, Golgi, lysosomes and late endosomes; oocytes are smaller than controls, stage 9-10 oocytes may display actin overgrowth and a frequently compromised membrane, stage 9-12 egg chambers frequently present a gap between the oocyte and the overlying follicle cells, vitellogenesis stage oocytes exhibit fewer and smaller vesicles/yolk granules; there are delaminating ring canals and intact ring canals leading into the oocyte show reduced amounts of membrane rather than the membrane thickening observed in controls. The frequency of multinucleate nurse cells is lower at 20[o]C than at 25[o]C. Most late stage egg chambers have defects in centripetal cell migration.
Female adults bearing Ykt6C clones show a mild rough eye phenotype and a double-bristle phenotype on the thorax.
