A genomic fragment containing 15.5kb beginning ~8kb upstream of the Clk translation start and ending ~2.5kb downstream of the Clk stop codon, in which 15/16 serines that act as phosphorylation sites have been replaced by alanine. The construct is tagged at the C-terminal end with Tag:V5.

Flies expressing Clk^15A.T:SV5\V5 in a Clk^out mutant background manifest generally weaker behavioral rhythms that are approximately 1.5 hours shorter compared to their wild type controls following exposure to entrainment conditions (12 hours light: 12 hours dark followed by several days in constant darkness). Clk^15A.T:SV5\V5, Clk^out mutant flies fail to entrain correctly to daily temperature cycles in the absence of light, but entrainment does occur in the presence of light.
