Description

Clkout is a 5398bp deletion generated through FLP-induced recombination between PBac{WH}Clkf06808 and PBac{WH}f03095. The remaining PBac insertion has been excised. The Clk transcription start site and regulatory sequences that direct spatial and circadian expression have been deleted.

Clkout homozygous adults reared under constant 25[o]C temperature and entrained to 12h:12h light:dark cycles show an increase in dark phase activity, show a decrease in morning startled activity, but do not show an obvious effect in evening startled activity, as compared to controls; upon transfer to constant darkness for 7 days, these individuals do not show circadian rhythmicity and almost do not show peaks of activities, as compared to controls.

Clkout homozygotes bearing either ClkTag:V5.WT or ClkΔ657-707.Tag:V5 kept under constant darkness and subject to 12h:12h 24[o] C:29 [o]C temperature cycles exhibit bimodal peaks of activity with an increase in anticipatory activity before temperature transitions; these individuals maintain some rhythmicity and manifest 23.1h and 24.9h periods, respectively.

Clkout/Clkout flies lose circadian rhythms (0% show rhythmicity) under dark conditions.

Flies expressing Clk15A.T:SV5\V5 in a Clkout mutant background manifest generally weaker behavioral rhythms that are approximately 1.5 hours shorter compared to their wild type controls following exposure to entrainment conditions (12 hours light: 12 hours dark followed by several days in constant darkness). Clk15A.T:SV5\V5, Clkout mutant flies fail to entrain correctly to daily temperature cycles in the absence of light, but entrainment does occur in the presence of light.

Homozygous Clkout mutant flies are viable and show no obvious developmental defects.

Flies expressing ClkS258A.T:SV5\V5 in a Clkout mutant background exhibit locomotor activity rhythm defects. A small, but significant, lengthening of the free-running period is seen compared to controls.

Flies expressing ClkS912A.T:SV5\V5 in a Clkout mutant background exhibit locomotor activity rhythm defects. A small, but significant, lengthening of the free-running period is seen compared to controls.

Flies expressing ClkS525A.T:SV5\V5 in a Clkout mutant background exhibit locomotor activity rhythm defects. A small, but significant, reduction in the free-running period is seen compared to controls.

Flies expressing ClkS859A.T:SV5\V5 in a Clkout mutant background exhibit locomotor activity rhythm defects. A small, but significant, reduction in the free-running period is seen compared to controls.

ClkT:SV5\V5,T:Avic\GFP-EGFP significantly rescues circadian rhythms (94% of flies show rhythmicity with a 24.5hr period) in Clkout/Clkout flies under dark conditions.
