tbrd-2HR mutant spermatids are produced, but contain nuclei of various sizes and irregular mitochondria, and display aberrant cytokinesis at the onion stage. Primary spermatocyte morphology at earlier stages is also unaffected.
No chromosome defects are observed in G2 phase chromosomes of tbrd-2HR mutant testes. However at prophase I when chromatid starts to condense into the typical three main chromosome masses, the two prominent asters of microtubules present in wild type spermatocytes are either not formed , aberrant in aspect or present in excessive numbers. In the subsequent metaphase I. anaphase I and telophase I sister chromatid segregation appears abnormal.
tbrd-2HR1 is rescued by tbrd-2EGFP,Tag:polyHis
Expression of tbrd-2T:Avic\GFP-EGFP,T:Zzzz\His6 rescues the sterility and spermatid defects seen in tbrd-2HR mutant males.
