The Trh promoter region is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 32[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.Trh and Hsap\RELAAD.R50H05 in adult females significantly increases food intake of sated flies, compared to controls; sated flies also have a significant increase in proboscis extension reflex in response to sucrose and significantly increased preference for D-glucose over L-glucose, compared to controls.
