Description

Insertion of a Mi{Hto} element in the last intron of Poxn ( 2R:15 ,829,437 (+) -Release 6). The hostile takeover (hto) exon in the Mi{Hto} element, which contains a T:Zzzz\FLAG tag and the mCherry form of Disc\RFP, splices to the last Poxn exon. This results in a fusion protein being expressed under the control of Scer\UAS regulatory sequences in the Mi{Hto} element.

Detailed Description

Expression of one copy of PoxnBRB under the control of Scer\GAL4GMR.PU produces flies that have pale/mottled and glazed (lacking clear separation of ommatidia) eyes.

Expression of PoxnBRB under the control of Scer\GAL4Bx-MS1096 results in the production of extra vein material, especially between L3 and L4 and near L5.

Expression of PoxnBRB under the control of Scer\GAL4pnr.PU severely impairs thorax development and eliminates abdominal pigment bands. Although the thorax does not expand properly, the number of macrochaetae in the Scer\GAL4pnr.PU-expressing zone of the thorax roughly doubles.

Expression of PoxnBRB under the control of Scer\GAL4ptc-559.1 results in failure of the femur and tibia to elongate in pupal stage flies.

Co-expression of PoxnBRB and ctBRO under the control of Scer\GAL4GMR.PU produces an eye phenotype that is qualitatively very similar to when either transgene is expressed alone. This phenotype is not suppressed upon co-expression of ctJF03304.

Expression of ctJF03304 does not suppress the wing phenotypes seen when PoxnBRB is expressed under the control of Scer\GAL4Bx-MS1096.

Comments

Expression of PoxnJF02136 almost completely rescues the eye phenotypes seen when PoxnBRB is expressed under the control of Scer\GAL4GMR.PU, although the posterior region remains mispatterned.
