ouib29/ouib74 mutant embryos complete embryogenesis, hatch normally and show no apparent morphological defects after hatching. They arrest development in the first instar larval stage and never molt into second instars. All ouib29/ouib74 mutant larvae die by 144 hours after egg laying. The majority of heterozygous animals become pupae.
The ecdysteroid titer in ouib29/ouib74 mutant animals is significantly reduced. When fed yeast paste containing 20E after hatching they are able to molt to the second instar larval stage or later.
When ouib29/ouib74 mutants are fed yeast paste supplemented with cholesterol or 7-dehydrocholesterol (7dC), the larvae still arrest at the first instar larval stage. The larval arrest phenotype is rescued when the animals are fed yeast paste supplemented with 5β-ketodiol (5βkd).
ouib74/Df(3R)ED5330 mutant animals arrest at the first instar larval stage but are able to reach the second instar stage when fed yeast paste containing 20-hydroxyecdysone (20E).
ouib29/ouib74 has lethal - all die during first instar larval stage phenotype, suppressible by spoUAS.Tag:HA/Scer\GAL4phtm.PU
Expression of spoScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4phm.PU suppresses the first instar larval arrest phenotype seen in ouib29/ouib74 mutants.
