UAS regulatory sequences drive expression of exons 4-7 of Nmnat with a DsRed(Unk) tag inserted in exon 5 and an AcGFP (AcGFP1) tag inserted in exon 6. This construct will produce two alternative transcripts: one containing the DsRed(Unk) sequence (proxy for splicing to RB) and one containing the AcGFP1 sequence (proxy for splicing to RA).
Flies with expression of Nmnat4-7.AltReport.Scer\UAS.T:Disc\RFP,T:Acoe\GFP driven in neurons by Scer\GAL4elav-C155 that are subjected to heat shock stress shift towards splicing RB; when expression is driven in glia by Scer\GAL4repo.PU, splicing is driven towards RA; ubiquitous expression driven by Scer\GAL4Act.PU leads to an overall increase in both splicing forms (RA and RB) in the brain.
