Description

UAS regulatory sequences drive expression of a Drosophila codon-optimized E(spl)m8-HLH open reading frame (no 5'UTR or 3' UTR sequences are present) that carries mutations in the CK2 phosphorylation site (three Ser to Ala replacements). The open reading frame is tagged at the N-terminal end with Tag:FLAG.

At stage 11 expression of E(spl)m8-HLHCK2.UAS.Tag:FLAG under the control of Scer\GAL4da.G32 leads to a reduction in neuroblast numbers when compared to control embryos.

Expression of E(spl)m8-HLHCK2.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR42G10 in embryos results in significant reduction of thoracic neuroblast daughter cell proliferation at stage 12 (but not stage 14), thoracic neuroblasts at stage 12 (but not stage 14), abdominal neuroblast daughter cell proliferation at stage 12 and 14, but no significant difference in abdominal neuroblast proliferation at stage 12 and 14, and also display reduced cell number of the neuroblast NB5-6T lineage, as compared to controls.
