The wild-type Hsap\TPI1 coding sequence is expressed under the control of Tpi regulatory sequences at the endogenous Tpi locus (the human coding sequence replaces the endogenous Drosophila coding sequence). The Hsap\TPI1 coding sequence has been modified to ensure expression in Drosophila (the sequence has been recoded for Drosophila codon usage and has the Drosophila intron-exon and splicing architecture). It was inserted into the Drosophila genome via phiC31:int-mediated integration into the attP site present in TpiattP.
Hsap\TPI1Tpi.WT, Tpisgk-js10/TpihTPI.M82T has long lived phenotype
One copy of Hsap\TPI1Tpi.WT (in which the coding sequence of endogenous Tpi is replaced by the human ortologue sequence) in an effectively Tpi-null background (i.e. TpihTPI.WT/Tpisgk-js10) leads to adult exhibiting a small increase in lifespan, but to no obvious defects in bang sensitivity or heat sensitivity, as compared to controls.
