Insertion of sequence encoding termination codons flanking a w+* marker. This results in a truncated protein containing the first 353 amino acids of wild-type Bsg25D. The mutant protein lacks the Smc domain and all but one of the coiled domains.
Ends-out gene targeting was used to replace a segment of the Bsg25D gene after aa 353 with a mini-white gene.
majority die during embryonic stage
Bsg25DN/Bsg25DN embryos exhibit a significantly decreased hatching rate, as compared to wild type, but no significant decrease in pole cell numbers.
