Expression of StripdsRNA.shRNA.Scer\UAS.9 under the control of Scer\GAL4RapGAP1-OK6 results in a significant increase in the number of satellite boutons on neuromuscular junctions (NMJ) in third instar larval 6/7 muscles (this increase is also observed in larvae expressing both StripdsRNA.shRNA.Scer\UAS.9 and StripdsRNA.shRNA.Scer\UAS); the total avg. number of boutons/NMJ as well as the muscle area is unchanged compared to controls. The amplitude of both evoked and spontaneous miniature excitatory junction potentials is unaffected but the frequency of mEJPs is significantly increased in larvae expressing StripdsRNA.shRNA.Scer\UAS.9 compared to controls.

Expression of StripdsRNA.shRNA.Scer\UAS.9 under the control of Scer\GAL4elav.PU leads to altered actin organization in neuromuscular junctions in third instar larvae.

The increased number of satellite boutons on neuromuscular junctions in third instar larvae expressing StripdsRNA.shRNA.Scer\UAS.9 under the control of Scer\GAL4RapGAP1-OK6 is not further enhanced by co-expression of either mtsJF02805 or CkaHM05138 but is suppressed by combination with a single copy of wtsx1 or co-expression of wtsJF02741 RNAi, it cannot however be suppressed by co-expression of ykiScer\UAS.cHa. The satellite bouton number phenotype is rescued by combination with single copy of hpo42-47, whereas the increased frequency of miniature excitatory junction potentials characteristic for StripdsRNA.shRNA.Scer\UAS.9 expressing larvae remains high. The increased bouton number is also suppressed by co-expression of enaS187A.Scer\UAS but not enaS187D.Scer\UAS.
