The expression of Sec71DN.Scer\UAS under the combined control of Scer\GAL4GSG2295 and RU486 treatment from mid-third instar larval stage onwards leads to dorsal multidendritic ddaC neurons exhibiting a fully penetrant defect in dendrite pruning 16h-24h after puparium formation, as shown by the persistence of some dendritic branches, but exhibiting no obvious defects in dendritic arborization at the white pupa stage, and leads to no defects in the elimination of ddaF neurons by apoptosis at 16h after puparium formation, as compared to controls; at wandering third instar larval stage, this expression leads to dorsal multidendritic ddaC neurons exhibiting Golgi apparatus defects, as shown by the severe decrease in the number of GalT or GM130 punctate structures in the soma, and exhibiting decreased protein secretion, but exhibiting no obvious defects in ER exit sites, as shown by the apparently normal pattern of Sec31 punctate structures in the soma, as compared to controls.
Clonal expression under the control of Scer\GAL4109(2)80 leads to dorsal multidendritic ddaD/E neurons exhibiting defective pruning, as shown by the persistence of some dendrites at 19h after puparium formation, as compared to controls.
