2153bp deletion that removes the entire first exon and part of the first intron of Graf. Generated by imprecise excision of P{EP}GrafG275.

In the Graf1/Df(1)BSC756 adult brain, mushroom body Β-axon fibers overextend beyond the midline, such that the Β lobes from both hemispheres appear to fuse; this phenotype starts being seen during the pupal stage, from 36h APF onward. Graf1 hemizygous adult males show a similar phenotype; in addition they exhibit a significant decrease in long-term memory (in aversive olfactory learning assays).
Graf1 homozygous mushroom body Β cell clones overextend beyond the midline.
Homozygous Graf1 mutants are viable and fertile.
Graf1/Df(1)BSC756 or Graf1/Y mutant third instar larvae show much higher levels of circulating and total (i.e. circulating plus sessile) hemocytes and also have significantly more hemocytes in the primary lymph gland lobe. This is attributable to increased production of plasmatocytes as the levels of neither circulating or sessile crystal cells nor the percentage of lamellocytes is changed relative to controls and it is also just the plasmatocyte population that is selectively increased in the lymph gland. This plasmatocyte overabundance appears to be primarily due to increased proliferation as the number of mitotically active hemocytes is significantly increased in both second and third instar larvae.
Primary hemocytes from Graf1/Df(1)BSC756 larvae show fluid-phase uptake deficiency and strongly reduced internalization of sSpi-GFP protein (spi encodes the principal ligand of the EGFR pathway) at high sSpi-GFP conditions (treatment with 30 ng/ml sSpi-GFP) but not at low sSpi-GFP conditions (treatment with 4 ng/ml sSpi-GFP), as compared to controls.

The increased concentration of circulating hemocytes characteristic for Graf1/Y mutant third instar larvae is suppressible by combination with a single copy of Egfrf24, Ras85De2F or spi1 and partially so with rl1; combination with rl1/rl10 reduces the hemocyte levels in Graf1/Y larvae to below wild-type levels, comparable to those seen in rl1/rl10 single mutants.

The increased number of total hemocytes (i.e. circulating plus sessile) observed in Graf1/Df(1)BSC756 mutant third instar larvae is fully rescued by expression of GrafScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4crq.PO in the mutant background. Scer\GAL4crq.PO-driven expression of GrafScer\UAS.T:Ivir\HA1 also rescues the fluid-phase uptake deficiency characteristic for primary hemocytes isolated from Graf1/Df(1)BSC756 larvae.
