Reported amino acid change:

K10term

Comment:

The mutation is reported as R (AAG) to stop (TAG) at amino acid 10. Codon 10 is AAG but this encodes Lys (K) so it should have been reported as K10stop. Surrounding sequences show that the mutation is in amino acid 10.

shiXFRT2 clones in the third instar larval fat body fail to induce autophagy upon starvation, as shown by their failure to increase LysoTracker levels. In agreement, under starvation conditions the clones show an accumulation and enlargement of early and late endosomes (identified as GFP-FYVE and GFP-Rab7 cytoplasmic puncta, respectively), an enlargement of lysosomes (identified as LAMP-GFP puncta) and their accumulation at the cell membrane, and an accumulation and enlargement of autophagosomes (identified as GFP-Atg8 cytoplasmic puncta and p62 cytoplasmic puncta), as compared to starved controls.
