The Q repeat region lacks secondary structure due to the insertion of Q to P mutations. The protein is tagged at the N-terminal end with Tag:HA and at the C-terminal end with Tag:MYC.
Expressing Hsap\ATXN3tr.Q70.pQp.UAS.Tag:HA,Tag:MYC under the control of Scer\GAL4ppk.PU has no impact on the dendrite morphology of class IV dendritic arborizing neurons in third instar larvae regardless of transgene expression level. Hsap\ATXN3tr.Q70.pQp.UAS.Tag:HA,Tag:MYC proteins fail to accumulate in the nucleus and are mostly found in the cytoplasm.
Expressing Hsap\ATXN3tr.Q70.pQp.UAS.Tag:HA,Tag:MYC under the control of Scer\GAL4109(2)80 has no impact on the actin cytoskeleton in dendrites of dendritic arborizing neurons in third instar larvae. Third instar larvae display similar motility and frequency of head turning to wild-type controls.
Expressing Hsap\ATXN3tr.Q70.pQp.UAS.Tag:HA,Tag:MYC under the control of Scer\GAL4221 has no effect on dendrite morphology in class I dendritic arborizing neurons.
