Description

Nucleotides 2340-2348 of the endogenous yrt gene have been mutated from TTCTTCTGG to CGTTTCCGT, resulting in the amino acid replacements F281R and W283R. This combination of mutations has been shown to abolish yrt oligomerization in vitro.

yrt⁷⁵ maternal mutant stage 11 and 13 embryos that carry a paternal yrt^F281R.W283R allele show epithelial polarity defects, as shown by the mislocalization of the apical Crb protein to the lateral membrane. This is accompanied by severe epithelial tissue defects, as shown by lack of head cuticle and a large hole in the dorsal cuticle.
