Most of the Vps39 coding sequence has been deleted (deletion coordinates 3R:18273942..18272027 ), allowing for expression of only the first 120 amino acid residues. A loxP cassette containing the Disc\RFPDsRed(T1).3xP3.cHa marker has been inserted in place of the deleted sequence.
1916bp of the Vps39 coding sequence has been deleted and replaced with a loxP cassette containing the Disc\RFPDsRed(T1).3xP3.cHa marker.
Vps39Δ1/Df(3R)Exel6178 mutant larvae exhibit trafficking defects in the brain and Garland cells. In the brain, the lysosomal marker Lamp mislocalizes to cytoplasmic granules. In the Garland cells there are enlarged enlarged vacuoles and postendocytic trafficking defects (e.g. endocytozed proteins fail to be efficiently incorporated into Lysotracker-positive autolysosomes), as compared to controls.
