Most of the Gr59c coding region has been deleted by homologous recombination. Sequence encoding the Scer\GAL4 driver, plus a w+mW.hs marker, has been inserted in place of the deleted sequence. Position of deletion endpoints on reference sequence determined from reported primer sequences.

Gr59c1 mutant I-a sensilla do not show the expected responsiveness to the bitter chemicals LOB, BER or DEN observed in controls, show ectopic responsiveness to CAF and UMB, and show the same lack of responsiveness to STR and SOA as controls; S-a sensilla show increased responsiveness to STR, but no changes in responsiveness to the other chemicals, as compared to controls; S-b sensilla show increased responsiveness to CAF, decreased responsiveness to BER, but no changes in responsiveness to other chemicals, as compared to controls; and I-b sensilla show no changes in responsiveness to any of the chemicals, as compared to controls. This correlates with decreased food avoidance to LOB, BER and DEN in two-way choice assays, as compared to controls.

The expression of Gr59cUAS.cUa under the control of Scer\GAL4Gr33a-GAL4 in Gr59c1 mutants rescues their decreased I-a sensilla responsiveness of to the bitter chemicals LOB, BER and DEN, and even induces hyper-responsiveness to LOB, BER and DEN in I-b, S-a and S-b sensilla.

The expression of Gr22eUAS.cSa under the control of Scer\GAL4Gr33a-GAL4 in Gr59c1 mutants rescues their decreased I-a sensilla responsiveness to LOB, BER and DEN, and even induces hyper-responsiveness to LOB and STR in both I-a and I-b sensilla, ectopic responsiveness to LOB, BER, DEN and STR in I-b sensilla, and hyper-responsiveness in S-a and S-b sensilla to all four chemicals.

The decreased responsiveness of S-a sensilla to LOB and DEN and the decreased responsiveness of S-b sensilla to LOB and DEN observed in Gr22eLL05566 mutants are partially suppressed by Gr59c1; the decreased responsiveness of S-b sensilla to BER is not affected by Gr59c1. The lack of responsiveness of I-a sensilla observed in Gr59c1 mutants is not suppressed by Gr22eLL05566.

The decreased food avoidance to the bitter chemicals LOB, BER and DEN observed in Gr59c1 mutants is enhanced by Gr22eLL05566.
