The endogenous Rab3 locus has been modified so that exon 2 is tagged at the N-terminal end with two copies of the Tag:HA epitope, and this tagged exon is inverted relative to the rest of the transcription unit, preventing expression of Rab3 protein. The inverted exon is surrounded by two pairs of incompatible recombinase target sites (FRT and FRT3) which are oriented such that the inverted exon can be stably inverted by expression of FLP recombinase (an odd number of inversions followed by an excision results in the exon being in the correct orientation to be expressed and being flanked by a single FRT3 and a single FRT site, which cannot recombine with each other).
